Correlation of the Genetic Map and the Endonuclease Site Map of Bacillus subtilis Bacteriophage SP02.

By marker rescue of bacteriophage SP02 sus mutants with purified bacteriophage SP02 DNA fragments, 11 of the 17 known bacteriophage SP02 sus loci were assigned to discrete DNA fragments. The left-most genetic locus, susA, was found to reside near one bacteriophage SP02 terminus (EcoRI-C1 fragment), whereas the right-most genetic locus, susP, was found to reside near the other bacteriophage SP02 terminus (EcoRI-C2 fragment). The physical locations of the intervening genetic loci were found to be consistent with the previously determined genetic order. Evidence was also obtained which suggested that at least one end of a transforming DNA fragment is degraded during DNA uptake by the competent bacterium.

Suppressor-sensitive mutants of Bacillus subtilis phage SPO2 were isolated and classified into 18 cistrons. A linear genetic map was constructed. The genes controlling early functions of phage development were clustered near the CI gene which may produce phage repressor.

Relationship between transformation in Bacillus subtilis and infection by bacteriophage SP02.

When bacteriophage SP02 infects a Bacillus subtilis culture during or shortly after transformation, the frequency of transformants among the resulting lysogens is greatly reduced relative to that in the uninfected culture. Characterization of temperate bacteriophages of Bacillus subtilis by the restriction endonuclease EcoRI: evidence for three different temperate bacteriophages.

Temperate bacteriophages of Bacillus subtilis were characterized according to host range and digestion of the bacteriophage genome by endonuclease EcoRI. The three bacteriophages, phi3T, SPO2, and phi105, were all heteroimmune, and the DNA digests showed dissimilar patterns by agarose-ethidium bromide gel electrophoresis.
